Predicting the Order of Arrival of the First Rejuvenation Therapies

As we approach the possibility of rejuvenation therapies, we see progress in the science of aging. These treatments will target the root causes of age-related diseases, ultimately managing aging as a chronic condition. From clearing senescent cells to restoring a youthful gut microbiome, each therapy aims to address specific molecular damages in the body.

1) Clearance of Senescent Cells
Multiple senolytic therapies are currently under development to remove senescent cells, which contribute to age-related degeneration. As studies show promising results in animals, the future holds new approved drugs and potential off-label usage for older individuals. The dasatinib and quercetin combination is showing rejuvenating effects, even though long-term human trials are needed for conclusive evidence.

2) Restoration of a Youthful Gut Microbiome
The gut microbiome plays a crucial role in overall health, and rejuvenating it can have lasting benefits. Fecal microbiota transplants and immunization with flagellin are promising techniques, with animal studies supporting their rejuvenating effects. However, data on human long-term health outcomes are still pending.

3) Clearance of the First Few Types of Amyloid
Therapies targeting amyloid proteins associated with age-related disorders, such as Alzheimer’s and heart disease, are making progress. While some immunotherapies effectively clear amyloid-β in the brain, further research is needed to improve side-effect profiles and expand treatment options. With continued advancements in this area, more therapies targeting different forms of amyloid may emerge in the near future.
