Mimicking Signaling from Exercise and Fasting in One Molecule

The metabolic responses to exercise and calorie restriction are vast, presenting numerous opportunities for drug development. Researchers explore a molecule that mimics the effects of key circulating signal molecules involved in regulating responses to exercise and fasting. This innovative approach aims to capture more benefits from exercise and fasting, highlighting the potential for new therapeutic interventions.


Elevating plasma levels of (S)-lactate (Lac) and/or (R)-beta-hydroxybutyrate (BHB) through strenuous exercise and prolonged fasting holds promising physiological effects. To harness these benefits, researchers introduce LaKe, a molecular hybrid of (S)-lactate and (R)-1,3-butanediol, which can be easily synthesized and converted into Lac and BHB. In animal studies, oral ingestion of LaKe led to increased plasma levels of Lac and BHB, triggering expected physiological responses such as reduced lipolysis and elevated levels of appetite-suppressing compounds.
